Green Mile. Firstly, when the barsteward doesn't wet the sponge and then, of course, John Coffey's execution.
 
The bit in American Psycho where Christian Bale is going at it with 2 hookers with Sussudio playing, followed by this bit

 
The matrix- Neo comes back to life (spoiler!!!)

Leon- Where he hands him the pin over (breathtakingly brilliant!)

Every film has one.... One defining moment.... I'm watching American History X at the moment..... The scene where he makes him bite the curb... Wow.

One of the most memorable bits from any film I've seen, pretty brutal. Great film though, one of my all time favourites.
